Assessment
of the Escherichia coli Tat Protein Translocation Systemwith Fluorescent
Proteins

Abstract The possibility of using
fluorescent proteins as probes to study the twin-arginine translocation (Tat)
system was assessed in Escherichia coli. When fused to the twin-arginine signal
peptide of trimethylamine N-oxide reductase, the DsRed2 red fluorescent protein
from the Discosoma sp. was successfully synthesized and folded in E. coli
cells. However, RR-DsRed2 aggregated inside the cells. Therefore, although
DsRed2 has been engineered from DsRed for faster maturation and lower
non-specific aggregation, it is still not compatible with Tat-dependent
translocation. In contrast, the jellyfish green fluorescent protein (GFP) was
efficiently exported into periplasm even when the RR motif was changed to KR or
RK. These results show that GFP can be used as an efficient reporter protein to
study Tat system, but DsRed2 is not suitable for such purpose because of its
aggregation property. In addition, when the protein concentration was similar,
the fluorescence intensity of KR-GFP and RK-GFP decreased compared with RR-GFP,
which would suggest that the twin-arginine signal peptide is not only essential
for mediating protein translocation, but also important for the folding of
down-stream protein.

利用荧光蛋白对大肠杆菌蛋白质Tat转运系统的研究

摘要 探讨了荧光蛋白作为报告蛋白用于蛋白质转运系统研究的可行性, 结果表明海葵红色荧光蛋白聚集在细胞质内, 不能转运至周质空间。 而水母绿色荧光蛋白在Tat信号肽和Tat转运酶的共同作用下,
以折叠形式转运至周质空间。 通过荧光定量分析表明信号肽保守序列中的双精氨酸是保证绿色荧光蛋白转运及转运效率所必需的, 且第二个精氨酸比第一个精氨酸更为重要。 同时,
揭示了Tat信号肽需要一定的高级结构才能行使功能; Tat信号肽不仅引导蛋白质的转运, 而且也参与蛋白质的折叠。
因此, 绿色荧光蛋白是非常理想的报告蛋白, 可用于研究Tat系统,
但是海葵红色荧光蛋白易于聚集而不适合于此目的。

细菌蛋白质Tat转运系统是1998年在大肠杆菌中发现的一种蛋白质转运系统, 因被转运的底物信号肽上含有双精氨酸(RR)保守序列核心S/T-R-R-x-F-L-K[1~9], 所以称为Tat
(twin-arginine translocation)转运系统。 这些底物是一些与细菌厌氧呼吸和细菌分裂有关的酶或蛋白质, 所以Tat转运系统与细菌的生命活动有着十分密切的关系。 底物蛋白除含有双精氨酸信号肽(Tat信号肽)这一特性外, 还以折叠的形式转运,
未折叠的线状蛋白质则通过Sec转运系统。 大肠杆菌Tat转运系统的功能单位Tat转运酶由三种Tat蛋白(TatA、
TatB 和TatC)组成。 蛋白质序列分析表明Tat蛋白都是穿膜蛋白, 经免疫共沉淀实验证实了TatA、 TatB和TatC共存于细胞膜上, 分离了TatAB和TatABC复合体[10~12]。 并在体外反应体系中组装了具有活性的TatABC复合体[13]。
TatB和TatC是Tat转运酶的重要成分,
近来的研究还表明TatC的氨基酸种类和分布对转运功能有着重要的影响[14~16]。 这些研究进展对Tat转运系统的进一步研究无疑有很大的促进作用。
自Tat转运系统发现以来, 多以天然依靠Tat转运系统转运的酶作为报告蛋白, 如大肠杆菌的三甲胺基-N-氧化物还原酶[TMAO (trimethylamine N-oxide) reductase,
TMAO还原酶]、
氢化酶-2(hydrogenase-2)等厌氧呼吸酶。 一般通过酶活性或蛋白质印迹等方式来测定酶的转运与否和转运的多少, 但这些指标会受酶活性测定条件及测定的敏感性等的影响。 为了建立更为直接、
方便的检测手段, 尝试用海葵红色荧光蛋白(DsRed2)和水母绿色荧光蛋白(GFP)作为报告蛋白, 将TMAO还原酶的信号肽(TorA-RR)基因与荧光蛋白基因重组来研究细菌蛋白质Tat转运系统。 由于荧光蛋白正确折叠后才能形成荧光[17], 所以可根据荧光的有无和分布确定荧光蛋白的折叠和转运状况; 通过定量测定荧光的强弱确定Tat转运系统的转运效率,
从而明确影响转运的因素。
本研究表明,
连接在TorA-RR信号肽下游的红色荧光蛋白在大肠杆菌中虽可正确折叠后产生红色荧光, 但不能转运至周质空间。 经同样处理的绿色荧光蛋白可正确折叠, 并以折叠形式在Tat蛋白的参与下转运至周质空间。 进一步以赖氨酸(K)不同程度地取代信号肽中的精氨酸形成新的信号肽(TorA-KR, TorA-RK和TorA-KK), 以研究精氨酸对绿色荧光蛋白转运的影响。
发现转运效率与取代的精氨酸位置有一定的相关性。 揭示精氨酸是绿色荧光蛋白高效率转运所必需的, 而且第二个精氨酸比第一个精氨酸更重要。
这种一级结构的改变导致介导转运功能的降低或丧失, 说明信号肽一级结构的变化可能影响高级结构的形成及被Tat系统有效地识别, 进而影响其功能。 同时, 显示Tat信号肽对蛋白质折叠和转运都有至关重要的作用。
1 材料和方法(Materials
and Methods)
1.1 材料
1.1.1 菌株和质粒 实验菌株TG1(Δ(lac-pro)supE
thi hsdΔ5/ F’traD36 proA+B+ lacZΔM15)、
MC4100A(F-Δ(argF-lac)U169 araD139 rpsL150
thi flb5301 deoC1 ptsF25 relA1 ara+)、
B1LKOA(如同MC4100A ΔtatC)、
BODA(如同MC4100A ΔtatB)、
ELV16A(如同MC4100A ΔtatA)和CU164A(如同MC4100A secY39cs, zhd-33∷Tn10)[18,19]。
质粒p-DsRed2(ampR, DsRed2)购自Clontech公司、
pRR-DsRed2(ampR, DsRed2克隆于pRR-gfp)、 pgfpmut2(ampR, gfp gene)[17]、 pRR-colV (ampR, cvaC克隆于pBAD8730)[18]、 pRR-gfp (ampR, gfp克隆于pRR-colV)、
pRKgfp、 pKRgfp、 pKKgfp、
pAR-gfp[18,19]。
1.1.2 培养基 采用Luria-Bertani (LB)培养基, 必要时添加氨苄青霉素(100
mg/L)(Amp)和阿拉伯糖(2 g/L)(APA)或葡萄糖(2 g/L)(APG)。
1.1.3 主要化学试剂 限制性内切酶、 T4连接酶、 聚合酶等购自Roche Molecular Biochemicals公司。
1.2 方法
1.2.1 培养条件 前培养取单一菌落接种于4~5
mL LB或APG培养基中, 37 ℃、
120 r/min振荡培养过夜或至A600为0.6时,
100倍稀释后进行扩大培养。
1.2.2 抗阿拉伯糖菌株的筛选 将待处理菌涂布于含曙红次甲基蓝(EMB)和阿拉伯糖的LB平板培养基上, 37 ℃过夜培养,
挑取紫红色的单菌落, 分离纯化。
1.2.3 克隆荧光蛋白基因 为克隆绿色荧光蛋白基因, 设计引物为:
gfp up (5′-aag aag gag ata taa cat gca
gca aag gag-3′), gfp down (5′-tga cca tga agc ttg cat gcc tgc-3′), 并在上下游分别引入了NheI,
pstI和HindIII酶切位点。 为克隆红色荧光蛋白基因, 设计了引物: DsRed2 up (5′-tcg cca cca tgg cta gct ccg aga acg
tca-3′), DsRed2 down (5′-cta tta ggc ttg act gca gac aag ttg
gta-3′), 并在上下游分别引入了NheI, NotI酶切位点。 以上引物均由MWG-Biotech France公司合成。 采用Expand high fidelity PCR
system (Roche Molecular Biochemicals)在梯度PCR合成仪合成。 酶切鉴定扩增片段。
1.2.4 扩增片段的基因克隆和序列分析 绿色荧光蛋白基因PCR产物经纯化后, 用NheI和HindIII酶切后克隆到质粒pRR-colV相应位点。 红色荧光蛋白基因PCR产物经纯化后, 用NheI和NotI酶切后克隆到质粒pRR-gfp相应位点, 连接转化TG1后, 在APA平板上培养过夜, 在紫外灯下选择发荧光的菌落, 提取质粒, 酶切鉴定, DNA序列测定由MWG-Biotech
France公司测定。
1.2.5 pRR-gfp和pRR-DsRed2转化 转化pRR-gfp和pRR-DsRed2到MC4100A及各tat基因缺陷株B1LKOA、
BODA、 ELV16A和SecY基因缺陷株CU164A中。
显微镜观察荧光分布并用CCD相机(Color Cool Viw, Photomic Sciences)摄像并处理(Image Pro-Plus software)。
1.2.6 细胞各组分的制备 细胞培养如前, 添加0.2%阿拉伯糖诱导荧光蛋白基因表达。 收获细胞采用溶菌酶-EDTA-冰水渗透压处理[3], 获得细胞周质空间成分,
再用French Press法得到细胞质成分。
1.2.7 电泳 10%聚丙烯酰胺凝胶电泳40 mA、
40 min。
1.2.8 定量测定荧光蛋白的强度 制备细胞周质空间和细胞质成分, 经活性凝胶电泳后, 定量测定细胞不同组分中的荧光量(Spex Fluorolog III 荧光仪)。
2 结果(Results)
2.1 抗阿拉伯糖菌株的筛选
因为torA-gfp重组基因的表达受控于阿拉伯糖启动子, 但araD缺陷菌株分解阿拉伯糖会产生有害的中间代谢物。
在EMB-阿拉伯糖平板上筛选了抗阿拉伯糖代谢的紫红色MC4100A、 B1LKOA、
CU164A、 BODA和ELV16A菌株。 各菌株在含阿拉伯糖的培养基上生长正常。
2.2 荧光蛋白基因扩增产物的克隆及序列分析
海葵红色荧光蛋白基因和水母绿色荧光蛋白基因的PCR扩增产物经酶切后与相应载体连接, 经氨苄青霉素抗性和表型筛选后, 分别提取质粒并酶切鉴定, 得到二个与相应荧光蛋白基因片段大小相似的片段(0.69 kb和0.73
kb), 序列测定的结果进一步说明目的基因已克隆于载体中。 重组质粒分别命名为pRR-DsRed2和pRR-gfp。
2.3 荧光蛋白在不同细胞中的分布
将所得的重组质粒pRR-gfp和pRR-DsRed2分别转化MC4100A、 CU164A、
B1LKOA、 BODA和ELV16A各菌,
得到了一系列的转化子。 经纯化分离,
培养后提取质粒, 分别用NheI和HindIII酶切、 NheI和NotI酶切鉴定, 确定含有相应的荧光蛋白基因。
经阿拉伯糖诱导培养, 荧光蛋白基因在各菌株中均能表达。 在MC4100A/pRR-DsRed2和B1LKOA/pRR-DsRed2细胞中只能观察到红色荧光点(图1)。
此结果表明虽然DsRed2溶解性较DsRed有所提高,
但是红色荧光蛋白仍聚集在细胞内不能被输送出去。 与此相反,
在野生型菌MC4100A/pRR-gfp细胞外围有一明显的绿色荧光光环, 而在tat基因突变株ELV16A/pRR-gfp(ΔtatA)、
BODA/pRR-gfp(ΔtatB)和B1LKOA/pRR-gfp(ΔtatC)中,
绿色荧光只是均匀分布在整个细胞中, 且细胞成对或呈短链相连不分离(图2)。

2.4 绿色荧光蛋白的转运
制备细胞周质空间和细胞质提取液, 进行活性聚丙烯酰胺凝胶电泳。 结果显示MC4100A和CU164A菌株的细胞周质空间提取液有绿色荧光蛋白条带; 但对于B1LKOA和BODA菌株只在细胞质提取液中有荧光蛋白条带, 在周质空间提取液中没有荧光蛋白条带(图3)。 在细胞质提取液中绿色荧光蛋白是以带有信号肽的前体形式存在, 而在细胞周质空间中由于绿色荧光蛋白转运后信号肽被切除, 所以绿色荧光蛋白的分子大小有差异。

2.5 信号肽中双精氨酸对绿色荧光蛋白转运的影响
通过荧光分光光度计测定细胞质内(图4, 白色块带)和周质空间中(黑色块带)的绿色荧光强度, 结果表明取代了保守序列中的不同双精氨酸形成的信号肽TorA-KK, TorA-KR和TorA-RK降低了荧光蛋白的转运效率。
第二位精氨酸的取代比第一位精氨酸的取代的负作用大, 完全取代则不介导绿色荧光蛋白的转运。 此外,
TorA-KR, TorA-RK和TorA-KK总体荧光量较TorA-RR总体荧光量逐渐有所下降。

3 讨论(Discussion)
3.1 作为细菌Tat转运系统报告蛋白的外源荧光蛋白性质上有差异
通过实验证明不同的外源荧光蛋白由于其自身性质的差异, 在作为细菌Tat转运系统的报告蛋白时表现出不同的结果。 在野生型MC4100A和CU164A菌株中,
绿色荧光蛋白可以转运至周质空间以成熟形式存在[图3(A), (B)]。 而在各tat基因缺陷株B1LKOA和BODA中绿色荧光蛋白仅以前体的形式存在于细胞质中, 没有转运到周质空间[图3(C), (D)], 这说明绿色荧光蛋白在大肠杆菌中可正确折叠, 并在tatB和tatC基因产物的共同参与下转运, 与Sec系统无关。 因而可作为Tat转运系统的报告蛋白[19]。 海葵红色荧光蛋白虽可正确折叠发出红色荧光, 但不能转运。 活性凝胶电泳实验表明, 在MC4100A菌株和tat基因缺陷株细胞质抽提液中均有红色荧光蛋白条带, 但周质空间抽提液中无红色荧光蛋白条带(图未显示), 这一结果与显微镜观察结果一致。
绿色和红色荧光蛋白序列具有23%同一性, 分子大小近似(分子量分别为27 kD和28
kD), 三维空间结构相同[20]。
尤其重要的是两者荧光基团都是在蛋白折叠后, 由3个氨基酸通过自我催化氧化反应形成环状结构而产生的。 绿色荧光蛋白以单体形式存在, 而红色荧光蛋白则为四聚体并易于聚集[21], 以致于在胞内聚合形成大分子而不能转运而积聚在细胞质中。 从而也说明了转运通道的大小是一定的, 不允许太大的分子通过。 故红色荧光蛋白不宜作为报告蛋白。
3.2 双精氨酸是绿色荧光蛋白高效率转运所必需的
早期的研究表明Tat信号肽的双精氨酸对蛋白质转运起着决定性的作用, 绝大多数情况下不可取代[22,23]。 用改变的信号肽TorA-RK, TorA-KR和 TorA-KK介导绿色荧光蛋白的转运来研究双精氨酸在转运中的作用, TorA-KR的转运效率高于TorA-RK,
表明信号肽中第二个精氨酸比第一个精氨酸重要。 TorA-KK不能介导绿色荧光蛋白转运, 表明精氨酸对信号肽的功能有极大的影响。
这说明了精氨酸可能不仅通过其极性基团与Tat转运酶相互作用, 还必须形成一定的高级结构才能发挥信号肽的作用。
3.3 信号肽不仅引导蛋白质的转运还影响蛋白质的折叠
水母绿色荧光蛋白本身是无信号肽的折叠蛋白, 在水母和大肠杆菌中它可正确折叠, 说明绿色荧光蛋白自身含有蛋白质折叠的信息。 本研究表明结合于绿色荧光蛋白上的野生型Tat信号肽并不影响下游绿色荧光蛋白的折叠, 但突变体TorA-KR、
TorA-RK和TorA-KK总体荧光量较野生型TorA-RR总体荧光量逐渐有所降低。
此外, 结合于Sec转运系统的信号肽则使下游绿色荧光蛋白不能正确折叠[24], 这些提示存在于折叠蛋白质外部的信号肽对折叠有着显著的作用。 Tat信号肽和Sec信号肽都具有三个相同的区域, 但两者的氨基酸组成有着明显的差异[25], 因而可推测信号肽上的氨基酸组成对蛋白质折叠有直接的影响。 已有文献证明,
存在于折叠蛋白质外部的前导肽对蛋白质的折叠确实有一定的影响[26]。
所以蛋白质的折叠信息并不仅仅存在于自身的一级结构中, 与外部信息亦有一定的关系。
利用水母绿色荧光蛋白作为报告蛋白的研究结果显示, Tat信号肽中的双精氨酸对于蛋白质的高效率转运是必需的, 尤其是第二位精氨酸;
信号肽作用的发挥极有可能与具一定的高级结构有关, 其功能不仅引导蛋白质转运还参与蛋白质的折叠。
